약물 타겟 억제제를 찾기 위해 X-ray를 활용하여 생체 내 단백질의 삼차원 구조를 연구하는 실험실입니다. 약물 개발에 있어 필수적인 단백질의 정제 및 단백질-단백질, 단백질-약물 상호 작용을 구조적으로 조사하여 약물 스크리닝을 수행합니다. 유전자 조작, 세포 배양, 단백질 정제 및 분석 분야에서 다양한 경험을 쌓아 취업 시 다양한 분야에 지원이 가능한 장점이 있습니다. 최신 장비들을 보유하고 있어 실험자들이 다양한 데이터를 얻을 수 있으며, CD, MALDI-TOF, ITC, DSC, DLS, Mosquito, Monolith, MALS, RT-PCR, AKTA와 같은 장비를 자유롭게 활용할 수 있는 실험실입니다.
Our group at Chungbuk National University is dedicated to unraveling the structure-function relationships of protein-protein or protein-RNA complexes, with a particular focus on nuclear transport-related protein complexes, including the importin family. Eukaryotic cells are compartmentalized into the nucleus and cytoplasm by the nuclear envelope, with intracellular signals transmitted through the continuous and bidirectional transport of proteins between these compartments via nuclear pores. To understand the molecular mechanisms underlying such intracellular communication at the atomic level, we crystallize and solve the structures of nuclear transport carrier proteins.
The main research areas include structural studies on selective binding of export cargoes by Exportin-5, characterization of a new nuclear transport receptor Hikeshi, investigation of selective nuclear import of the C2H2 zinc-finger protein Snail by importin β, biosynthesis of tunicamycin antibiotics, elucidation of the selective nuclear export mechanism of small RNAs, high-resolution structural analysis of the pre-microRNA nuclear export machinery, conformational changes in tryptophan synthase from a hyperthermophile upon complex formation, dissociation of nuclear import complex by RanGTP, structural basis of sugar-recognizing ubiquitin ligase, and structure of importin-beta bound to SREBP-2.
Our laboratory primarily employs X-ray crystallography to investigate the three-dimensional structures and interactions of proteins within the body to identify potential drug targets. We perform structural analyses of key proteins and their interactions with drugs to facilitate drug screening. With expertise in genetic manipulation, cell culture, protein purification, and analysis, our laboratory provides versatile skills applicable across various scientific disciplines. Equipped with state-of-the-art instruments such as CD, MALDI-TOF, ITC, DSC, DLS, Mosquito, Monolith, MALS, RT-PCR, and AKTA, our laboratory facilitates comprehensive experimentation and data acquisition.
